The first international forum on relaxin was a workshop held in Hawaii in 1980. This was followed by the Kroc Foundation conference in Santa Ynes, California in 1981 and then the First International Congress on the Hormone Relaxin in Florence in 1982. After a hiatus of several years, the Second International Congress on the Hormone Relaxin was held in Adelaide in 1994. Since that time, the enormous advances in molecular biology and protein chemistry have led to a stellar growth in relaxin research. Novel sites of action of the hormone have been elucidated, numerous new mammalian primary structures determined, more detailed mechanisms of action postulated, the control of relaxin secretion better understood, non-pregnant roles of the hormone identified, and, importantly, relaxin-related peptides (particularly INSL 3 and 5) identified and possible synergistic actions with relaxin proposed. A raft of novel clinical applications of relaxin has also been explored. Together, these exciting developments made the time ripe for a third international conference. We were therefore delighted to welcome many colleagues, old and new, at Relaxin 2000, the first international forum on relaxin and related peptides in the new Millennium. This conference, which we hope will now be held every four to five years, was held at the Cable Beach Inter-Continental Resort at the delightful old pearling town of Broome in the far north-west of Australia from 22-27 October, 2000.

This Pediatric Nephrology series is a focus on salient points which at the time of each annual seminar are of importance to the practicing pediatrician and nephrologist, the clinical researcher, and basic researcher interested in clinical problems. Hence the format of selected papers and panel discussions to capture the tenor of the times. More thorough coverage of many of the subjects can be found in current journals and textbooks listed in the authors' references. Those searching for the conventional should look there rather than here since our aim is not to cover each subject in its entirety but to secure attention to the controversial aspects of the subjects, dispel the notion that there is one answer to a question, and raise the level of inclination toward dynamic problem solving. The basic subject chosen this year reflects dominant concerns this year and the participants chosen--speakers and discussants--represent certain views relevant to the subject at this time. To reflect the tempo and flavor produced by this unique blend, the discussions are included almost verbatim. For some this means readability; for others, excess verbiage. The careful reader will notice that I have been the chairman of all sessions and have moderated all discussions. This is in keeping with our aim to ferret out interrelated basic questions and varying answers to the subjects--seen as related in problems and solutions. In the discussions, all names have been deleted.
As the number of drugs acting on calcium and potassium channels grows, there is a need for a continuous reappraisal of the cellular machinery controlling them. The present volume provides an update on the basic knowledge, the molecular targets of the two channels, and the importance the drugs that bind them have as pharmacological tools and therapeutic agents. This work was presented at the 6th International Symposium on PHARMACOLOGICAL CONTROL OF CALCIUM AND POTASSIUM HOMEOSTASIS: BIOLOGICAL, THERAPEUTICAL, AND CLINICAL ASPECTS, in Florence (Italy) on October 4-6, 1994. Because of the recent advances in the field, discussions on potassium channels were included for the first time. At least six classes of voltage-dependent calcium channels have been defined based on their physiological and pharmacological properties. Among them, L-type channels, mediating long lasting currents, are better characterized. Calcium homeostasis within the cell is not only regulated by calcium channels; intracellular calcium stores, in particular a pool contained in the lumen of specialized areas of the endoplasmic reticulum, are rapidly exchanged with the cytoplasm and playa key role in the control of calcium homeostasis. This area, however, has not yet been exploited from a therapeutic point of view. Potassium channels are present in virtually every cell type, excitable and nonexcitable, and are distinguished by structural, biophysical, and pharmacological criteria. Different classes including voltage-gated, ligand-gated, AP-sensitive, and G-protein coupled among others, have been defined by their primary regulatory and gating mechanisms.
